Deoxyribonuclease I, or DNase I is an endonuclease that acts on phosphodiester bonds adjacent to pyrimidines to produce polynucleotides with terminal 5′-phosphates. It cleaves preferentially double-stranded DNA and in the presence of Mg2+ single stranded DNA.
- Used to nick DNA as a first step to incorporate labelled bases into DNA
- Suitable for DNase footprinting
- Improves yield in primary cell isolation by removing extracellular DNA
- Decreases viscosity in protein samples by removal of DNA contamination
Unit definition: 1 Kunitz unit catalyzes an increase in absorption of 0.001 at 260 nm per minute at 25 °C, pH 5.0 when acting on highly polymerized calf thymus DNA (1).
