Amido Black Staining of transfer membranes
Solutions:
- Amido Black Staining solution (10x concentrate):
1 % (w/v) Amido Black dissolved in 40 % (v/v) Ethanol und 10 % (v/v) Glacial acetic acid - Destaining solution:
40 % (v/v) Ethanol, 10 % (v/v) Glacial acetic acid and 2 % (v/v) Glycerol
Staining protocol:
The blotting membrane is incubated for 5 minutes in amido black staining solution (1:10 dilution) and then destained in destaining solution.
Note: Amido black is not a reversible stain, but more sensitive in detection than Ponceau S, comparable to a Coomassie® Brilliant Blue R stain.
